Any one here is doing protein protein interaction work? I hope we can have a discussion on this topic one day.
As described in the paper i just uploaded (http://www.xinkexue.com/bib.php?do=ref&refid=4618)"This review is intended as a practical guide to the analysis of such protein-protein interactions. We discuss biochemical methods such as protein affinity chromatography, affinity blotting, coimmunoprecipitation, and cross-linking; molecular biological methods such as protein probing, the two-hybrid system, and phage display: and genetic methods such as the isolation of extragenic suppressors, synthetic mutants, and unlinked noncomplementing mutants. We next describe how binding affinities can be evaluated by techniques including protein affinity chromatography, sedimentation, gel filtration, fluorescence methods, solid-phase sampling of equilibrium solutions, and surface plasmon resonance. "
So any one who has some experience on this, we can have a discussion.
To JamesLZ. The method on the prediction of protein protein interaction site is mainly based on 3D structure. I noticed there are some sequence based prediction which are too complicated for me. Do you know any web sever predicting protein protein interaction site based on sequence but not structure? Many thanks! |
